Best way if you can afford it: Take a taxi for approx. $35. Time involved depends on traffic, but I'd say you're looking at 45 minutes at least.

Best way on a budget: AirTrain ($5) from the terminal to Howard Beach Station (Subway) or Jamaica Station (Long Island Railroad)

~then: Subway to Manhattan (Line A)- Approx. $2, around 45 min. plus wait time for the train to pick you up. At 42nd St., Change to Line 1 or 9, a few stops to 103rd St station.

~ or: Long Island Railroad to Penn Station ($12) - Approx 35 min. plus wait time. Ride, A, C, or E line to 42nd St., Change to Line 1 or 9, then go to 103rd St.

I've done the AirTrain/Subway thing once. I'm sticking to cabs. It's not *that* complicated to do it by train, but after a long flight, I'm just not in the mood for all the waiting and the transfers.

It's now a $45 flat-rate to anywhere in Manhattan, tolls nonwithstanding.

I always use New York Airport Service http://www.nyairportservice.com/

It's $15 from JFK to 3 different spots in NY (Grand Central, Port Authority, Penn Station). It takes a little time, but saves quite a bit of money if you're traveling by yourself.

Then you could just take the subway from Port Authority to West End Studios for $2/per person.

Also with a cab (and xanthous is right about the $45 flat rate and tolls not included) you have to figure in a 10-20% tip on top of that. It sure is faster and more comfortable to just jump in a cab and arrive at your destination without dragging your bags all around, but if you really want to save the extra cash, you can do the bus thing.

If they don't pick me up I ride a cab, to much hassle to have the bags on and be on the subway at the same time plus the drunken flights always get to you (by you I mean me) and yeah the fare is 45$.
